Blood Coagulation as Coagulation Dysregulation

Prior to human transplantation application, clarification and elucidation of the pig organ compatibility with the human host thromboregulation system are prerequisite in the pig-to-human or pig-to-primate xenotransplantation. Pig xenografts promote intensive thrombotic response of human blood, although pharmacological interventions and genetic targeting approaches have been made. To reduce acute thrombosis, human thromboregulatory genes have been transgenically introduced into pig donors, including endothelial protein C receptor (EPCR) and human CD36 (hCD36), and the α1,3Gal-T KO/hCD46/hEPCR pig has been generated. In addition, a triple KO pig donor also showed the decreased binding levels of human natural Abs in the acute antibody-dependent rejection.
