Enhancing the Prodrug ADME Profile: An Emerging Area to Overcome the Issues of Cancer Drug Resistance

Despite significant improvements in the management of different cancers, the emergence of cancer resistance accounts for the majority of chemotherapy failings. A diverse approach is required for an effective management of cancer, according to growing knowledge of the molecular complexity of cancer and causes of drug resistance as well as considerable clinical experience. Hence, cancer research aims to develop new methods to combat cancer drug resistance. In the recent decades, prodrugs have sought the attention of the researchers which focus on different aspects like enhanced ADME profiling of the drug, decreasing side effects, site-specific delivery, etc. Prodrugs offer a great therapeutic potential. They have offered some great benefit in different disorders like inflammation and Crohn’s disease. This book chapter covers the drug resistance mechanisms in chemotherapy at macro as well as micro level and efficiency of prodrug as a possible strategy to target drug resistance in numerous cancers.
