Immortality has been a longstanding aspiration of humanity. Despite many failure experiences in the attempts to achieve longevity, scientists have continued to delve into the study of aging. Over the decades, many scientific breakthroughs have been made and numerous theories have emerged to explain aging mechanisms, leading to proposals for various anti-aging interventions. Significant strides have been made from theoretical concepts to practical applications, showing promising results in slowing down aging and extending healthy lifespans. Effective methods recognized today to combat aging include caloric restriction, moderate exercise, and activation of autophagy. These interventions work by regulating processes such as DNA damage repair, protein homeostasis, mitochondrial quality control, telomere length maintenance, inflammation reduction, and oxidative stress homeostasis. One of the major achievements in nicotinamide coenzyme I (NAD+) research has been the discovery that it can promote healthy aging and even extend the lifespan of many organisms. The signaling pathway mediated by NAD+ is closely related to the signaling mechanisms involved in classical anti-aging measures. What’s more, previous research suggests that the aging process is highly malleable, and the promising results in animal studies have the potential to extend to humans. This has sparked enthusiasm in both basic research and translational applications on the anti-aging properties of NAD+.
