Phyto-parasitic nematodes (PPNs) are a significant threat to global agriculture due to their ability to cause substantial crop losses. Effective management of PPNs requires the development and implementation of accurate diagnostic tools and efficient management strategies. Molecular diagnostic tools have emerged as a promising approach to rapidly and accurately identify PPNs and gain insights into their genetic diversity and virulence. In this chapter, the current state of molecular diagnostic tools for PPNs and their application in the management of these pests is reviewed. An overview of the various molecular techniques used for the detection of PPNs, including polymerase chain reaction (PCR)-based techniques, DNA sequencing, and isothermal amplification techniques is provided. The advantages and limitations of each of these tools and case studies demonstrating their application for the identification of specific PPN species are also highlighted. Next, the various molecular techniques used for the management of PPNs, including RNA interference (RNAi), genome editing, and gene silencing are thoroughly discussed. Finally, the challenges and considerations (regulatory and ethical) associated with the development and implementation of molecular diagnostic and management tools for PPNs are highlighted. Molecular diagnostic and management tools have emerged as powerful approaches for the effective management of PPNs. Continued development and improvement of these tools will be critical for reducing the impact of PPNs on global agriculture. The integration of molecular techniques with traditional management strategies has the potential to provide more efficient and sustainable solutions to PPN management.
