Advanced Molecular Techniques in the Identification of Phytopathogenic Fungi

A great range of fungal pathogens infect plants, especially economically significant crops, and these infections lead to the most harmful illnesses and significant yield loss worldwide. There are estimated to be about 1.5 million fungi species. Among them, the number of described species is less than 5%. The global agricultural sector has significant economic challenges due to phytopathogenic fungal species, which resulted in a substantial drop in crop yield in both quality and quantity. These illnesses have a significant negative economic and production impact on the global agricultural sector. The most crucial phases in creating management plans for phytopathogenic fungi are detection and diagnosis. Because there are many different fungal pathogen species, races, biotypes, and strains within each species, and because crop cultivation practices vary from region to region, it is difficult to develop direct detection assays. Diagnostics for fungal plant diseases rely on a wide range of technologies, from conventional taxonomy to innovative molecular equipment. The potential of the organism to be cultured, the length of time involved, and the need for in-depth taxonomical knowledge are some of the major drawbacks of traditional approaches. Diseases must be diagnosed early and accurately to anticipate outbreaks and allow enough time to create mitigation techniques. Modern molecular techniques, such as DNA-based techniques, now offer crucial equipment for precise plant disease identification. Efficient amplification platforms, DNA barcoding, varied quantitative PCR, probe creation, and next-generation sequencing based on RNA sequence have revolutionized the study of fungus differentiation and diagnosis. Although molecular diagnosis techniques have made tremendous advancements over the past two decades, there are still more advancements that need to be developed and utilized by phyto-diagnosticians. Ultimately, molecular diagnosis methods employed in phyto-diagnostic clinics must be simple, reliable, effective, affordable, and user-friendly to compete with and more advanced than conventional methods.
