Utilizing HPLC for Efficient Metabolite Purification
摘要
High-performance liquid chromatography (HPLC) plays a pivotal role in efficient metabolite purification, a crucial step in metabolite profiling. This review delves into the significance of metabolites, exploring their role as cellular building blocks, fuel sources, and signaling molecules. The concept of the metabolome, a dynamic collection of metabolites within a biological system, is introduced. HPLC’s function in metabolite separation is explained, highlighting its analogy to a sorting station that segregates metabolites based on their unique properties. The integration of mass spectrometry (MS) with HPLC (HPLC-MS) is emphasized as a powerful technique for metabolite identification and quantification. The advantages of HPLC-MS, including its exceptional sensitivity, selectivity, and structural elucidation capabilities, are discussed. Furthermore, the abstract explores the diverse applications of HPLC-based metabolomics, encompassing disease biomarker discovery, drug development, environmental monitoring, and research on plants and microbes. Finally, the abstract acknowledges the value of complementary analytical techniques like NMR spectroscopy and underscores the importance of a multi-platform approach for comprehensive metabolite profiling and a deeper understanding of cellular biochemistry.
