Scaling Up Nature’s Chemistry: A Guide to Industrial Production of Valuable Metabolites
摘要
The exploration of metabolites and their diverse applications across food, pharmaceuticals, and industrial sectors are broadly discussed in this chapter. The chapter begins by elucidating the pivotal role of metabolites as indicators of an individual’s metabolic status, shaped by genetic predispositions and environmental influences. Emphasizing metabolomics as an evolving analytical discipline, it underscores its objective of detecting, quantifying, and dissecting primary and specialized metabolites within specific biological contexts. The discourse extends to the intricacies of large-scale metabolite production, addressing challenges spanning strain and metabolic engineering, regulatory compliance, environmental impact mitigation, and the inherent complexity of biological systems. Through emphasizing the imperative of advancements in these domains, it advocates for enhanced efficiency and sustainability in industrial metabolite production. Furthermore, the chapter delineates strategies for surmounting production hurdles, exemplified by the iterative Design–Build–Test–Learn (DBTL) cycle framework. It stresses the optimization of each cycle phase, encompassing genetic manipulation, phenotyping methodologies, data interpretation via computational tools, and knowledge integration for iterative improvement. Exploring diverse production platforms including bacterial, yeast, and fungal systems, the chapter underscores their respective merits in yielding valuable metabolites and facilitating industrial processes. Notably, bacterial systems are praised for their cost-effectiveness, sustainability, and genetic manipulation prowess, while yeast and fungal systems offer versatility and diversity. In summary, this chapter furnishes a comprehensive comprehension of metabolites, their multifaceted significance, and the associated challenges and strategies inherent in industrial-scale production. It serves as an invaluable resource for researchers, industry practitioners, and students invested in metabolomics and its myriad applications.
