摘要

Metabolomics is a rapidly advancing field that integrates bioinformatics and high-throughput analytical techniques to analyze metabolites quantitatively and qualitatively. These metabolites serve as intermediaries in metabolic reactions and provide insights into pathological mechanisms, enabling therapeutic intervention options. Metabolomics is particularly sensitive to environmental and genetic stimuli, making it crucial to consider various factors while conducting studies to ensure efficiency. Genome editing technologies, such as CRISPR-Cas9, TALENs and ZFNs, have revolutionized the field of genetics, particularly in cellular chemistry. These tools enable the identification, evaluation and construction of modified strains, making it possible to engineer industrially important strains. CRISPR-Cas9, in particular, has proven to be a powerful genetic editing tool, with its primary objective being to increase on-target efficiency and decrease off-target effects. Metabolic pathway engineering is essential for the production of various compounds, and it involves disrupting or overexpressing key genes. The process can be achieved by integrating exogenous pathways from other organisms or de novo-constructed pathways, or by instigating metabolic endogenous pathways. The core components of metabolic pathways include deletion of competing pathways, overexpression of bottleneck enzymes, selection of host organisms, computational systems, transcriptional control engineering, engineering of enzymes, discovery of novel synthetic gene editing tools and reduction of metabolic load. Precision medicine, which involves tailoring metabolic profiles for disease treatment, is an emerging approach that allows scientists and researchers to customize treatment options for patients based on their responsiveness to particular therapies and susceptibility to certain ailments. Metabolomics plays a crucial role in precision medicine by analyzing tiny molecules, particularly metabolic intermediates formed in biochemical reactions. This approach has been used to investigate interactions between protein downstream products, genes and environmental stimuli, as well as to ascertain the pathophysiology of different diseases like cancer and Type 1 diabetes mellitus. In conclusion, metabolomics and genome editing technologies have significantly advanced our understanding of metabolic reactions and their implications for disease mechanisms and treatment options. These fields have opened up new avenues for the development of personalized medicine and the production of various compounds through metabolic pathway engineering. However, it is crucial to consider various factors while conducting metabolomics studies and to ensure the specificity and affinity of genome editing tools for effective genomic modifications.
