Peptide-Based Vaccines in Cancer Immunotherapy

The field of cancer research has made impressive strides in recent years. Nevertheless, cancer continues to be a leading cause of death, and cancer metastasis is a problem that needs to be solved quickly because it is the primary cause of therapeutic failure. Unfortunately, the most widely used chemotherapy drugs are nonselective, can harm healthy tissues, and have side effects that have a significant negative impact on patients’ quality of life. Rapid developments in anticancer vaccination opened the door to the identification of several targets to induce an immune response against one or more tumor-associated antigens (TAAs). A tumor-reactive T-lymphocyte response is specifically required for tumor-specific immune activation. One of the really promising strategies that have just been granted clinical approval is the use of peptide-based vaccinations that target T cells. The therapeutic potential of peptide-based vaccinations made specifically for an individual is also being investigated. When used as immunotherapeutic agents, peptide-based substances have a lot of potential to treat a variety of malignant tumors. The development of peptide-based cancer vaccines and delivery methods has received a lot of attention because it mimics the functional domains of proteins that have very specialized immuno-regulatory roles or that regulate the fate of tumor cells. The connections between various peptides and immune systems are still not fully understood, though. The enthusiasm for peptide-based vaccination regimens in the context of cancer has been reignited by improvements in the engineering of peptides, adjuvants, and delivery methods, and a number of clinical trials based on the usage of peptides are being carried out by pharmaceutical companies. The obstacles to effective cancer immunotherapy are not specific to peptide-based vaccination regimens; they apply to all immunotherapeutic techniques. This chapter provides a thorough focus on the design, categories, and applications of peptide-based cancer vaccines, the challenges to effective immunotherapy, and will discuss potential options for the future. It will also describe the advances in the identification, design, and delivery of peptides, and the role of peptides in regulating the functions of the innate and adaptive immune systems.
