Cancer immunotherapy has transformed the landscape of oncology by exploiting the immune system’s capabilities to selectively target and eradicate cancer cells. Nevertheless, persistent challenges such as restricted immune evasion, tumor infiltration, and unintended off-target effects continue to impede progress. Polymeric nanoparticles (PNPs) have been developed as a promising solution for improving the precision and effectiveness of immunotherapeutic agents. This chapter examines recent breakthroughs in leveraging PNPs to optimize outcomes in cancer immunotherapy, exploring the PNPs design, synthesis, and functionalization for the targeted delivery of antigens, immune modulators, and various bioactive compounds. Furthermore, the chapter investigates how PNPs overcome limitations in immunotherapy, encompassing sustained release, immune activation, and modulation of the tumor microenvironment. By highlighting the preclinical and clinical advancements of PNPs in cancer immunotherapy, the chapter emphasizes their potential to transform the landscape of cancer treatment.
