Cell-free fetal DNA (cffDNA) has emerged as a transformative entity in prenatal diagnostics, originating from placental trophoblasts and circulating freely in maternal blood. Its discovery marked a pivotal moment, unlocking new dimensions in noninvasive prenatal testing. The clinical utility of cffDNA has undergone extensive exploration, establishing itself as a cornerstone in screening for chromosomal aneuploidies, monogenic disorders, and various fetal anomalies. However, challenges such as its relatively low concentration, ethical considerations, equitable access to healthcare resources, and cost-related concerns pose significant hurdles. Current technological advancements aim to address these challenges, holding the promise of enhancing the effectiveness and accessibility of cffDNA-based testing. This chapter explores into the existence, clinical applications, and the myriad challenges associated with cffDNA in detail.
