Polyunsaturated Fatty Acids and Traumatic Brain Injury

Polyunsaturated fatty acids (PUFA), including precursors and long-chain PUFAs (LC-PUFAs), are necessary fatty acids. Due to the inability of mammals to create precursors, they must be obtained from nutrition. They can be converted into LC-PUFAs. It is wise to take LC-PUFA supplements, which control cell and brain function, because humans have such a low conversion rate. Long-chain polyunsaturated fatty acids (LC-PUFAs) are characterized by their carbon chain length of 20 or more carbon atoms and multiple double bonds. Key members of this group include eicosapentaenoic acid (EPA, 20:5 ω-3), docosahexaenoic acid (DHA, 22:6 ω-3), and arachidonic acid (AA, 20:4 ω-6).The composition of LC-PUFAs is significant in both n-3 and n-6 series, with n-3 LC-PUFAs, such as EPA and DHA, being derived from dietary sources like fish oil, while n-6 LC-PUFAs, such as AA, are primarily obtained from vegetable oils. The synthesis of these fatty acids involves a series of enzymatic reactions, including desaturation and elongation, facilitated by specific enzymes such as delta-5 desaturase (D5D) and delta-6 desaturase (D6D). These enzymes introduce double bonds at specific positions in the carbon chain, which is essential for the formation of bioactive lipid mediators, including eicosanoids and specialized pro-resolving mediators (SPMs) that regulate inflammation and promote tissue repair.
