Bispecific Antibodies: An Emerging Concept in Antibody-Based Cancer Therapies

Bispecific antibody-based treatment options have emerged as an impending therapeutic strategy for various deadly cancers including several hematological malignancies and solid tumors. A bispecific antibody is a synthetic antibody construct that contains binding sites directed against different cellular and molecular targets. Its function and activity can be dependent on this bispecific linkage that would not normally be obtained by the presence of monospecific antibodies with the same specificities, or it can be an alternative for a monospecific antibody mixture. Bispecific antibodies have emerged as modern promising targeted therapy options that could combine different modes of actions in the same molecule potentially facilitating its development, approval, manufacturing, and clinical usage comparing to monospecific antibodies. Here, we have shed light on the basics of bispecific antibodies’ mode of action and their manufacturing. Next, we have provided current bispecific antibody treatment strategies in various preclinical and clinical models of solid and hematological cancers. Furthermore, we have included an updated clinical cancer trials details that employing various formats of bispecific antibodies.
