The Protein–Lipid Overlay Assay stands as a pivotal technique in the realm of molecular biology, serving as a fundamental tool for the study of protein–lipid interactions. Central to the intricate machinery of cellular processes, these interactions play crucial roles in various physiological functions, ranging from signal transduction and membrane trafficking to cellular metabolism and homeostasis. Protein–lipid interactions control the location, activation, and function of proteins, which is crucial for lipid metabolism, membrane trafficking, and cell signaling. Phase separation of lipids inside the membrane bilayer, driven by thermodynamic forces, produces highly organized subdomains known as lipid rafts inside the cell plasma membrane. The lipid raft areas exist as “raft-like” structures that float atop the otherwise fluid plasma membrane. The lipid raft’s functional involvement in intercellular and intracellular communication through lipid-lipid, lipid-protein, or protein-protein interactions is greatly influenced by the variations in lipid and protein composition within the raft. The regulation of target proteins’ conformational states at biological membranes by phosphoinositides may be facilitated through protein-protein interactions involving lipid-binding domains.
