Understanding the intricate interactions between proteins and lipids is crucial for deciphering cellular processes. This chapter explores advanced techniques that shed light on these interactions. Cross-linking coupled with mass spectrometry offers a powerful tool for identifying protein–lipid complexes and their stoichiometry. Fluorescence resonance energy transfer (FRET) imaging visualizes protein–lipid proximity in living cells. Nuclear magnetic resonance (NMR) provides high-resolution structural information on protein–lipid interactions at the atomic level. Electron paramagnetic resonance (EPR) spectroscopy, with its capability to study specific spin-labeled molecules, offers insights into protein dynamics within lipid environments. These complementary techniques, along with visualization methods like EPR imaging, provide a comprehensive toolbox for dissecting the dynamic interplay between proteins and lipids in biological membranes.
