A mutation refers to a change in the nucleic acid sequence within an organism’s genome. Mutations are essential for evolution to occur and play an important role in the origin of new species. Mutations are also driving forces for genetic diversity and adaptation. This chapter explores the significance of mutations and their assessment in the context of microbial technology. This chapter begins by introducing genes, gene functions, and phenotypes. Then we discuss the concept of mutations, their types, mechanisms, and factors influencing mutation occurrence. Next, we discuss the selection of mutants to enhance microbial productivity, strain improvement and its importance in microbial technology. This includes the methods and strategies employed to generate desired traits in industrially important microorganisms, including classical mutagenesis techniques, site-directed mutagenesis, and directed evolution approaches. In conclusion, understanding the mutations, their assessment methods, and employing effective selection strategies are essential for harnessing the full potential of microorganisms in various industrial applications and microbial technology.
