摘要

This chapter presents a comprehensive overview of messenger RNA (mRNA) technology, highlighting its historical evolution, underlying mechanisms, diverse platforms, manufacturing intricacies, and inherent advantages. Initially conceptualized within the framework of the central dogma of molecular biology in the 1950s–1960s, mRNA serves as an intermediary for genetic information translation into functional proteins. This chapter delves into the various mRNA platforms, encompassing conventional and self-amplifying mRNA types, elucidating their distinct attributes and applications. Manufacturing intricacies and the scalability of mRNA synthesis are discussed, outlining the crucial steps involved in production. Despite promising attributes, the technology faces challenges related to stability, immunogenicity, and delivery systems, which are analyzed in detail. Platform optimization strategies focusing on structural modifications, novel delivery systems, and adjuvant use are highlighted to enhance efficacy and stability, paving the way for future improvements. The narrative specifically addresses emerging opportunities in mRNA technology, emphasizing its pivotal role in combating infectious diseases, cancer, and metabolic disorders. This chapter further underscores the potential impact of mRNA technology in India, acknowledging the nation’s vast population and diverse health challenges. The discussion revolves around the need for infrastructure enhancement, cost-effective production, and skill development to harness mRNA technology’s potential effectively. Conclusively, this chapter provides insights into the transformative role of mRNA technology, outlining its promising future as a robust therapeutic and preventive tool. It underscores the imperative for continued research, collaborations, and technology refinement to address global health challenges and accelerate the integration of mRNA technology into clinical applications.
