Plants are diverse, and their interactions with living organisms make them more versatile. The plants respond to biotic and abiotic interactions through protein molecules. So, the expression of genes and formation of proteins are very important, and further regulation of genes is also vital in plant–microbe interactions. This book chapter addresses the multifaceted realm of plant proteomics, providing a comprehensive overview of the techniques, applications, and recent advancements in the study of plant proteins. It focuses on studying and expressing small peptides, like antimicrobial peptides, to complex proteins in heterologous expression systems and the related troubleshooting. By exploring the intricate network of plant proteomics, this chapter sheds light on the fundamentals of proteomics, outlining the various techniques employed in plant proteomic studies. From gel-based methods like two-dimensional gel electrophoresis (2-DE) to advanced mass spectrometry (MS)-based approaches, the readers gain insights into the diverse strategies employed to unravel the plant proteome.
