Dalbergia sissoo Roxb. ex DC is a valuable fast-growing tree native to the Indian subcontinent. It is widely recognised for its economic, ecological, and medicinal importance. However, overexploitation, diseases, and environmental stress have caused a significant decline in its population, necessitating effective conservation strategies. Tissue culture, a versatile biotechnology tool, has emerged as a promising approach for large-scale propagation and preservation of this species. This chapter comprehensively analyses the advances in tissue culture techniques for D. sissoo, including callus induction, somatic embryogenesis, organogenesis, and micropropagation. It discusses the challenges of microbial contamination, genetic recalcitrance, somaclonal variation, and acclimatisation while suggesting future directions, such as molecular approaches, to enhance propagation efficiency.
