Class V SLRP has two members, podocan and podocan-like protein-1. They bind with collagens just like the other members of SLRPs and although they have leucine-rich repeat domains, they differ in their C-terminal ends and are also considered to act as glycoproteins. This chapter discusses the interaction of class V SLRPs with different signalling pathways, including Wnt/β-catenin, TGF-β, and PI3K/AKT/mTOR where (P) podocan and podocan-like protein-1 have a regulatory effect, and podocan is a negative regulator of cell proliferation and migration, and it can bring these effects by downregulating Wnt/β-catenin signalling. Involvement of class V SLRPs in various diseases, including diabetic nephropathy and different types of cancers is discussed including (E) epigenetic regulation of podocan and podocan-like protein-1.
