Applications of Advanced ASOs, RNA Mimics, and Others in RNA-Based Therapeutics

The rapid advancement of RNA-based therapies has revolutionized the management of various diseases, including genetic conditions, cancers, and viral conditions. This chapter focuses on the innovative uses of antisense oligonucleotides (ASOs), RNA analogs, and other cutting-edge RNA technologies in therapeutic applications. ASOs have demonstrated great potential in modulating gene expression by targeting specific mRNA sequences, contributing to substantial therapeutic advancements in diseases like spinal muscular atrophy and Duchenne muscular dystrophy. This chapter offers a summary of the different types of ASOs, their mechanisms of action, and the clinical achievements that have solidified their role as a key element in RNA-based therapies. Additionally, RNA analogs such as locked nucleic acids (LNAs) and peptide nucleic acids (PNAs) have emerged as important assets in this area because of their enhanced stability and stronger binding affinity for target RNA sequences. These mimics are facilitating the development of more precise and effective gene modulation techniques, providing new hope for conditions that were once deemed untreatable. This chapter further examines the design, synthesis, and therapeutic applications of these RNA mimics, highlighting their role in advancing personalized medicine. Emerging RNA technologies, including CRISPR/Cas RNA-guided systems and RNA aptamers, are also discussed, showcasing their transformative potential in gene editing, targeted drug delivery, and biosensing. The integration of these technologies with traditional RNA therapeutics is poised to unlock new therapeutic avenues, offering synergistic effects and improving patient outcomes. This chapter concludes by addressing the challenges facing RNA-based therapeutics, particularly in terms of delivery, stability, and regulatory hurdles, while also providing insights into future directions for research and clinical application.
