Ribonucleic acid provides insights into active biological processes making it an indispensable tool for modern diagnostics. RNA reflects active gene expression, giving a dynamic view of the cellular state. RNAs are isolated from different clinical sample types using various approaches including organic extraction (TRIzol method). Through the medium of this book chapter, we have attempted a comprehensive and scientific examination of various tools, methods and techniques that aid in the diagnostic applications using RNA. RNA-based diagnostics harness the dynamic information provided by RNA to detect, monitor and understand a broad spectrum of diseases. Core amplification techniques like reverse transcription-polymerase chain reaction (RT-PCR), quantitative PCR, loop-mediated isothermal amplification, a substitute to qPCR making it suitable for point-of-care applications have been discussed. Additionally, the chapter details advanced RNA detection and profiling tools such as microarray, RNA sequencing and CRISPR employed for diagnostic purposes. The chapter provides an integrated view of these RNA-based methods in clinical diagnostics, paving the way for improved patient outcomes and precision healthcare solutions.
