Protein folding/unfolding is an intriguingly complex process that has been studied for decades now. Atomic force microscopy, optical tweezers, and magnetic tweezers grouped under the umbrella of single-molecule force spectroscopy have complemented our understanding of this complex process by applying mechanical force as a denaturant. The current chapter discusses our previous understanding of the “protein folding problem,” effect of force as a denaturant, and principles and working of the single-molecule force spectroscopy tools and novel insights they provided on the process of protein folding and unfolding.
