摘要

Colorectal cancer (CRC) is a prevalent intestinal condition with significant global health implications, marked by high incidence, morbidity, and mortality rates. The research underscores the critical role of gut microbiota in CRC development and progression, challenging the traditional view that genetic mutations solely drive the disease. The incidence of CRC varies globally, with higher rates in regions with a high Human Development Index (HDI), while lower rates are observed in less developed areas. This disparity is shifting as CRC cases rise in developing countries but stabilize or decline in wealthier nations. Currently, approximately 1.93 million new CRC cases are diagnosed annually, making it the second leading cause of cancer-related deaths, following lung cancer, with around 904,019 fatalities each year. Epidemiological projections suggest a significant increase in CRC cases, anticipating 3.2 million cases and 1.6 million deaths by 2040. The gut microbiota’s influence on CRC is profound, with imbalances linked to the disease’s onset and progression. Lifestyle factors such as diet, alcohol consumption, physical inactivity, and obesity are implicated in altering the gut microbiome, potentially increasing carcinogenic metabolites. The rising incidence of CRC, particularly among younger populations, and the higher mortality rate due to late diagnosis are concerning. The gut microbiota’s ability to modify epigenetic mechanisms like histone modification, non-coding RNAs, and DNA methylation highlights its importance in CRC pathogenesis. Maintaining gut homeostasis through a balanced microbiome is crucial for intestinal health and cancer prevention. Probiotics, such as Lactobacillus and Bifidobacterium, have shown promise in CRC prevention and treatment, with ongoing research focusing on microbiota manipulation as a therapeutic strategy.
