Actinobacteria constitute a diverse and widely distributed group of bacteria prevalent in both freshwater and marine ecosystems, where they play crucial roles in ecological processes such as nutrient cycling and symbiotic interactions. Freshwater actinobacteria, including genera such as Streptomyces and Nocardiopsis, are more abundant in lake sediments than in dynamic river environments, where they produce antimicrobial compounds effective against pathogens. Marine actinobacteria occupy a range of habitats, from surface waters to deep-sea sediments, including extreme environments, and exhibit extensive genetic and metabolic diversity, resulting in unique bioactive metabolites with pharmaceutical potential. These compounds are promising candidates for the development of antibiofilm, antibacterial, and anticancer therapies. Key challenges in this field include difficulties in culturing, activation of silent biosynthetic gene clusters, low yields, complex purification processes, and genomic complexities. Emerging solutions include integrated omics, genome mining, synthetic biology, co-cultivation, and heterologous expression to unlock novel metabolites. Cost-effective fermentation, scalable bioprocesses, and regulatory pathways are essential for industrial applications. Marine and freshwater actinobacteria are promising sources of novel therapeutic agents and biotechnological innovations, underscoring the need for continued research and innovative cultivation strategies to maximize their potential applications.
