Synthetic Biology Tools for Marine Actinobacteria

Marine actinobacteria represent a vast, yet underexplored, reservoir of bioactive natural products with immense potential for pharmaceutical and industrial applications. Despite their genetic richness, the majority of biosynthetic gene clusters (BGCs) in these organisms remain silent under standard laboratory conditions, limiting the discovery of novel metabolites. Recent advances in synthetic biology, encompassing genome mining, clustered regularly interspaced short palindromic repeats-Cas-based editing, modular cloning, and heterologous expression, have revolutionized our ability to decode and engineer these cryptic pathways. This chapter provides a comprehensive overview of synthetic biology tools tailored for marine actinobacteria, including bioinformatics platforms such as antibiotics and Secondary Metabolite Analysis Shell, Prediction Informatics for Secondary Metabolomes, Bacteriocin Genome Mining Tool, and DeepBGC for BGC prediction, as well as genetic toolkits for promoter engineering, DNA assembly, and the design of regulatory circuits. It highlights the integration of multi-omics approaches and machine learning in pathway optimization and discusses emerging marine-adapted chassis systems, such as Salinispora tropica, for scalable biotechnological applications. Finally, this chapter addresses the persistent challenges in transformation efficiency, vector stability, and cultivation of marine isolates, outlining future directions for establishing standardized synthetic biology platforms that harness the full biosynthetic potential of marine actinobacteria.
