Rapid and Accurate Information Reading-Out

摘要

Reading-out information from DNA requires sequencing technology for data processing and interpretation. This chapter will first lay out how to use Sanger sequencing, next-generation sequencing (NGS) as well as HiFi-based, and nanopore-based sequencing methods for digital data storage, which offer varying trade-offs between speed, accuracy, and scalability. Next, we will discuss the basics on accelerating and optimizing sample preparation, signal calibration, robust biochemical workflows, and random-access capabilities. We will then present the fundamentals of error-correction designs, redundancy strategies, and consensus-building techniques, enhancing the precision of data recovery. We will end up with a presentation of two examples using text and image data, and their benchmarking results, revealing practical and throughput considerations for the development of readout approaches. The chapter connects with writing-in methods and influences DNA nanostructures for storage architectures, providing state-of-the-art knowledge and insights into achieving rapid and accurate digital data reading-out.
