Cancer Signaling Pathways and Cancer Biology

Cancer arises from a complex interplay of genetic, epigenetic, and environmental factors that dysregulate key cellular signaling pathways. Aberrations in pathways such as PI3K/AKT/mTOR, RAS/MAPK, WNT/β-catenin, Notch, Hedgehog, and p53 not only drive oncogenesis but also sustain tumor progression, resistance to therapy, and metastasis. This chapter provides a comprehensive overview of the major oncogenic signaling networks implicated in diverse cancer types, emphasizing their biological roles, molecular mechanisms, and therapeutic vulnerabilities. Integrating insights from The Cancer Genome Atlas (TCGA) and recent literature, we explore how mutational landscapes converge on critical nodes within signaling cascades. Furthermore, the chapter investigates signaling crosstalk, feedback loops, and metabolic reprogramming, offering a systems-level understanding of tumor biology. Special attention is given to current therapeutic strategies, including targeted therapies, combination regimens, and novel inhibitors that exploit pathway dependencies. Lastly, we discuss future directions for precision oncology, highlighting the potential of systems biology and omics-driven approaches to identify robust biomarkers and rational drug targets. Through this integrative perspective, the chapter aims to bridge fundamental cancer biology with translational applications, providing a foundation for innovative research and improved clinical outcomes.
