Novel Treatment for Endometrial Cancer

The mainstay of advances in novel treatments for endometrial cancer is immune checkpoint inhibitors (ICIs). However, the objective response rate of ICI-related treatments is approximately 40% for endometrial cancer, and patients still require novel therapies. Ongoing and other latest phase II/III clinical trials include a selective MDM2 inhibitor; a selective inhibitor of XPO1; mTOR inhibitors; a monoclonal antibody directed against HER2; a selective TIE2 inhibitor; antibody-drug conjugates (ADCs) targeting TROP2, FRα, HER2, HER3, and TF; and an alkylating histone deacetylase inhibitor (HDACi). ADCs are expected to become primary candidates for new treatments for endometrial cancer. In addition, it is also noteworthy that progress has been made in both making efficient use of existing antiangiogenic agents and in developing new drugs targeting p53, which has been a long-standing goal since previous studies have indicated that p53 mutations occur in endometrial cancer. In the future, we also need to verify the introduction and application of cancer genome profiling using whole-genome sequencing and AI systems for treatment individualization.
