This chapter explores the advancements and applications of modern molecular diagnostics, focusing mainly on Loop-Mediated Isothermal Amplification (LAMP). Introduced in 2000, LAMP provides rapid, cost-effective, and sensitive nucleic acid amplification without the need for thermal cycling, making it ideal for diagnosing infectious diseases, detecting plant pathogens, and conducting point-of-care tests in resource-limited environments. The chapter also discusses related innovations such as circulating tumor DNA (ctDNA), which is revolutionizing cancer diagnosis through real-time, noninvasive monitoring of tumor changes. Liquid biopsy and exosome-based diagnostics are complementary methods for early detection and personalized therapies. Additionally, Recombinase Polymerase Amplification (RPA), another isothermal technique, shows potential for clinical and agricultural applications. Microbial genome profiling helps uncover microbial diversity and mutualism in plants and ecosystems, while bioimaging technology enhances our understanding of plant biology, aiding crop breeding and stress analysis. CRISPR-Cas-based diagnostics like SHERLOCK and DETECTR are transforming pathogen and genetic marker detection. Together, these developments are set to change diagnostic practices across various fields. The chapter ends with an overview of current challenges and future opportunities for implementing these technologies in health, agriculture, and environmental monitoring.
