Scientists have access to diverse enzyme sources for nucleic acid modification, cutting, and propagation. Understanding the activity of each enzyme is crucial for optimizing reaction conditions and selecting the best source for specific goals. Successful cloning depends on choosing the right enzymes and high-quality reagents. This chapter provides insights into the roles and specific functions of various enzymes, including topoisomerases, phosphatases, methylases, nucleases, polymerases, and ligases, used in molecular cloning. It highlights common enzymes from each group, describing their characteristics and mechanisms of action. The aim is to guide readers in selecting the most suitable enzyme sources and conditions for successful molecular cloning by emphasizing key features and limitations of each enzyme.
