Fundamental Methods in Molecular Biology

Molecular biology has become a pivotal component of modern science, significantly transforming fields such as medical diagnostics, biotechnology, forensics, and genetic research. Central to molecular biology are several core methods that enable researchers to isolate, analyze, and modify biomolecules, including DNA, RNA, and proteins. This review provides a comprehensive overview of key molecular biology techniques, focusing on their principles, applications, challenges, and recent advancements. Extracting DNA is crucial in molecular workflows, which involve cell breakdown, purification, and separation for applications such as polymerase chain reaction (PCR), cloning, gene expression, and sequencing. PCR (molecular photocopying), amplifies DNA exponentially, transforming diagnostics, forensics, and genetic engineering. This review covers standard PCR and advanced variants that improve specificity. Techniques such as agarose gel electrophoresis (AGE) and polyacrylamide gel electrophoresis (PAGE) separate nucleic acids and proteins by size, with AGE used for DNA/RNA fragments and PAGE for high-resolution separation of proteins and small nucleic acids. SDS-PAGE and formaldehyde gel electrophoresis enhance resolution for gene expression and diagnostics. Protein analysis reveals structure and interactions; methods preserve their integrity for characterization. Electroelution, combining electrophoresis and band retrieval, aids purification for spectrometry and blotting. These core techniques form the foundation of molecular biology research. This chapter aims to not only summarize their principles and wide range of applications but also examine key innovations and address technical limitations that continue to shape the development of molecular biology.
