Antiproliferative Natural Metabolites to Inhibit Penis and Testicle Tumor

Penile and testicular cancers, though relatively rare, represent significant oncological challenges due to late diagnosis, limited therapeutic options, and treatment-associated toxicities. Emerging evidence highlights the potential of bioactive natural metabolites as safer, multitargeted alternatives or adjuncts to conventional therapies. This chapter elucidates the anticancer mechanisms and experimental evidence of natural metabolites like cordycepin, curcumin, resveratrol, thymoquinone, and withaferin A, in relation to penile and testicular malignancies. These phytochemicals exert their effects through modulation of oxidative stress, inhibition of key oncogenic transcription factors (NF-κB, STAT3), and regulation of cell-cycle and apoptotic machinery (p21/p27, Bax/Bcl-2, caspases). Cordycepin from Cordyceps militaris induces mitochondrial apoptosis and suppresses metastasis via PI3K/Akt and MAPK pathways. Curcumin downregulates HPV oncoproteins and inflammatory mediators, while resveratrol restores mitochondrial integrity and enhances antioxidant defenses against testicular oxidative injury. Thymoquinone and withaferin A exhibit potent cytotoxicity through ROS generation, p53 activation, and inhibition of angiogenesis. Collectively, these findings support the therapeutic promise of natural metabolites as chemopreventive and chemotherapeutic agents targeting molecular pathways underlying male reproductive cancers.
