Working with Worms: Protocols and Some Practical Considerations for a C. elegans Laboratory

The nematode, Caenorhabditis elegans, is being used as a model organism in biological research for decades, and its popularity among researchers, as well as its utility spectrum, is still increasing. Easy and open availability of protocols for employing this worm for a variety of assays has contributed considerably towards its adoption by many laboratories across the globe. While the basic protocols for routine maintenance and handling of this worm have been in the public domain for a long time, new protocols keep being reported with time. Earlier use of this worm was mainly restricted to neurobiology labs; however, the last two or three decades have witnessed use of wild type and transgenic strains of C. elegans for a wider spectrum of assays in labs working in areas as varied as diabetes, neurodegeneration, aging and longevity, wound healing, microbial infections, etc. The need for designing and validating new protocols arises as and when new applications for this worm model are envisaged. This chapter presents some protocols for the use of this worm in microbiological as well as non-microbiological assays, along with relevant troubleshooting tips.
