The dot blot and Western blot methods are techniques for specific detection of proteins of interest in a mixture. In the dot blot process, these proteins of a suspension are directly spotted onto a protein-adsorbing membrane. Using the Western blot technique, protein suspensions will be separated by gel electrophoresis in a carrier matrix for detailed and differentiated analyses according to size. The protein pattern in the gel matrix is transferred to a solid carrier membrane. After saturation and blocking of free and unbound surfaces in the carrier matrix, proteins are detected selectively by immunostainingImmunostaining. Proteins of interest are identified by binding to specific antibodies for detection followed by visualization of the signals. This chapter introduces the technique of gel electrophoresis and the subsequent analytics using Western blot and dot blot methods.
