Circulating Tumor Cells: Isolation and Molecular Characterization

The malignancy of tumors is characterized, among other things, by the progression of the primary tumor, its invasive growth, and the ability to metastasize. A significant parameterParameter for metastasis is the detachment of epithelial tumor cells from the cell association of the primary tumor and their circulation in the bloodstream. The analysis of circulating tumor cells (CTC) is of high diagnostic and prognostic importance. The membrane-bound form of the heat shock protein 70 (mHsp70), which occurs exclusively on malignantMalignant cells of almost all tumor entities, opens up a possibility to selectively and efficiently isolate CTCs from the peripheral bloodPeripheral blood, even after epithelial-mesenchymal transition. This chapter describes an innovative method in which polystyrene beads, immobilized with a monoclonal antibody directed against mHsp70, are used for the isolation of CTCs.
