Two-photon microscopy (2 PM) is a modern imaging technology that enables the observation of dynamic cellular processes deep within the tissues and organs of living organisms. The use of a pulsed ultrafast infrared laser allows high light penetration into the tissue under investigation. As a result, this imaging technology is well suited for a wide range of in vivo applications. This chapter briefly introduces the biophysical basics of 2 PM and illustrates its current applications in intravital imaging.
