Magnesium is an essential mineral that functions as a cofactor in over 300 enzymatic reactions, but remains a frequent deficiency state due to inadequate dietary intake and faulty absorption. The pharmacological rationale for multi-form magnesium supplementation to enhance systemic bioavailability, reduce gastrointestinal side effects, and sustain tissue-specific physiological processes is the subject of this review. Magnesium Breakthrough™, a seven-magnesium complex supplement, is a model of evidence-based formulation. This paper highlights using solubility assays, model GI digestion, and Caco-2 cell transport models in the supplement industry to measure absorption properties. It also suggests a novel approach in building a synergistic formula composed of different forms of the same element due to their unique characteristics. Based on a thorough literature review, it is seen that organic structures such as bisglycinate, malate, and Sucrosomial® magnesium demonstrate better tolerability and solubility, while threonate, taurate, and orotate may provide benefits for neurological function, cardiovascular function, and mitochondrial health. Together, the outcomes support a multi-form strategy as a coordinated strategy for optimizing magnesium delivery, enhancing physiological action, and achieving long-term compliance.
