Nanocarriers and Smart Delivery Systems for Enhancing Bioefficacy of Mushroom Compounds

The bioactives isolated from mushrooms explicitly have broader therapeutic potential, such as immunomodulatory, antioxidant, anticancer, anti-inflammatory, and neuroprotective properties. However, low bioavailability and rapid metabolism of mushroom bioactives often constrain their clinical application. Advanced nanocarrier-based drug delivery systems overcome these challenges by enhancing solubility, permeability, pharmacokinetics, and targeted drug delivery, thus offering a promising strategy for mushroom bioactives delivery. The various nanocarriers’ drug delivery systems, including polymeric nanoparticles, lipid-based carriers, and inorganic nanocarriers, for facilitating controlled drug release at the targeted site are covered in this chapter. Additionally, stimuli‑responsive nanocarriers—engineered to release bioactive compounds in response to specific triggers such as pH variation, temperature change, or enzyme activity—hold strong promise for enhancing the targeted delivery and therapeutic efficacy of mushroom‑derived compounds. The preclinical evidences like in vitro, and in vivo models for enhanced bioavailability and therapeutic efficacy are briefly discussed. Furthermore, the translation of the preclinical to the clinical sector emphasizes the real-world applicability of nanocarrier-based mushroom bioactives for immunotherapy, neuroprotection, and anticancer research. Also, this chapter comprehensively discusses the integration of mushroom-based nanocarriers into the pharmaceuticals and nutraceutical industries and commercial sectors, beyond their therapeutic implications. The regulatory compliances, safety evaluation, and global standards related to mushroom-based nanoformulations are critically assessed to enhance their success rate in commercial and clinical sectors. Overall, this chapter fills the gaps by connecting preclinical insights to clinical applications and unraveling the complete therapeutic potential of mushroom bioactive via advanced nanotechnology-based drug delivery systems. The future prospects for mushroom‑based therapeutics are promising, with continued advances in nanocarrier and smart delivery systems expected to further improve the stability, targeted delivery, and clinical efficacy of mushroom bioactives. .
