Inflammasome is a multi-protein complex that govern the maturation of the pro-inflammatory cytokines such as interleukin-1β (IL-1β) and IL-18 in response to a variety of intracellular stimuli. Recent discoveries have demonstrated that inflammasomes are involved in carcinogenesis and tumor progression. As a result, inflammasome has become an attractive target in the treatment of cancer. This chapter elaborates an overview of the biological roles of inflammasomes. Subsequently, we delineate the pivotal roles of the inflammasomes in carcinogenesis and tumor progression, especially in the context of solid tumors. After that, we discuss the approaches and pharmacological agents based on the inhibition of inflammasomes in the context of clinical immuno-oncology.
