Post-translational modifications (PTMs) provide regulatory mechanisms that allow for diverse and complex multifaceted cellular signaling, including phenotypic modulation. Small molecules that inhibit such PTM can provide novel therapeutic options to treat many diseases (including cancer). This chapter will focus primarily on inhibitors of acetylation but will also discuss the potential for targeting more recently discovered PTMs as crotonylation, butyrylation, and propionylation.
