Receptor tyrosine kinases (RTKs) are crucial in regulating cellular growth and division. When ligands bind to them, they initiate a series of intracellular signaling events, primarily through the Ras/MAPK pathway, which activate genes essential for progressing through the cell cycle. Abnormalities in this mechanism are typical in various cancers, where excessive RTK activation is often observed, underscoring the critical importance of RTK signaling in maintaining normal cellular growth and division. Here, we emphasize the role of RTKs in mitochondrial dynamics and functions. They engage different pathways to affect mitochondrial dynamics in cancer, as EGFR and some of its family members translocate to mitochondria and significantly impact disease severity and chemotherapy resistance.
