Receptor tyrosine kinases (RTKs), a family of transmembrane proteins, have drawn significant attention in cellular signaling due to their important roles in regulating cell growth, differentiation, metabolism, and survival. These receptors are key mediators in signal transduction pathways by responding to extracellular ligands. Dysregulation or aberrant signaling induced by RTKs (such as EGFR, AXL, and VEGFR) has profoundly contributed to a variety of diseases, including cancer. In cancer, abnormal RTK signaling is mediated through various mechanisms such as gene amplification, gain-of-function mutation, chromosomal rearrangements, autocrine activation, and loss of negative feedback. This chapter aims to present a thorough understanding of RTK concerning cancer. We have discussed inhibitors of RTKs and mechanisms of therapeutic resistance toward therapies.
