Topical Antineoplastics in Dermatology

Topical antineoplastic agents have been effective against various cutaneous lesions for multiple decades and serve as useful therapeutic tools in clinical dermatology [1]. Of note, efficacy of topical antineoplastics against basal cell carcinoma and actinic keratosis has been well acknowledged [2, 3]. Beyond these applications, certain antineoplastic agents have shown utility in treating multiple skin conditions outside of their FDA-approved indications. For instance, imiquimod has been used to treat various cutaneous conditions such as mycosis fungoides and Kaposi’s sarcoma, despite it being only FDA approved for perianal warts, basal cell carcinoma, and actinic keratosis [4]. Topical antineoplastics also have various mechanisms of action, from 5-fluorouracil, which inhibits metabolism, to imiquimod, which modulates the immune system [1, 5, 6]. With diversity in clinical applications and mechanism of action, topical antineoplastics have significant potential for future research regarding clinical efficacy or new compounds under development [7].
