Platelet-rich fibrin (PRF) is an autologous biological product derived from whole blood containing various growth factors, including bone morphogenetic protein (BMP), platelet-derived growth factor (PDGF), vascular endothelial growth factor (VEGF) acting as reservoirs of growth factors. Several platelet-derived components have been developed including fresh and exudate forms of PRF, however, the product is limited by its fast degradation time and restrained usage. Lyophilization is an alternative approach to increase the lifetime of bioproducts without changing the protein nature. This study aims to investigate the fabrication process, chemical and biological properties of different PRFs including human lyophilized PRF (Ly-PRF) and lyophilized PRF exudate (Ly-PRFE). Healthy volunteers are selected to collect whole human blood. The morphological and chemical composition of the obtained samples are characterized by scanning electron microscopy and Fourier-transform infrared spectroscopy, respectively. The kinetics of growth factors released from Ly-PRF and Ly-PRFE are also evaluated by PDGF-AB enzyme-linked immunosorbent assay (ELISA). Subsequently, cytotoxicity of all products is assessed using L929 cell line. ELISA results show that Ly-PRFE exhibits higher PDGF-AB content than Ly-PRF. Cell viability test demonstrates that both samples have no cytotoxicity effect and have great potential for tissue regeneration.
