Determination of Inhibitory Activity of Angiotensin-Converting Enzyme (ACE) and Carbohydrate Metabolism Enzymes (α-Amylase, α-Glucosidase, DPP-IV) from Leaves Extracts of Barringtonia Acutangula

The Barringtonia acutangula leaves contain compounds such as flavonoids, saponins, steroids, and tannins, which can reduce blood pressure and support diabetes treatment. This study aimed to investigate the inhibitory activities against the enzymes α-glucosidase, α-amylase, dipeptidyl peptidase-IV (DPP-IV), and angiotensin-converting enzyme (ACE) in vitro using all solvent extracts from leaves of B. acutangula. The powdered material from the leaves was extracted using 96% ethanol via the filtrated extraction method to obtain the total ethanol extract (referred to as TP extract). A portion of the total extract was then mixed with water and sequentially extracted with solvents of increasing polarity, including n-hexane, ethyl acetate, n-butanol, and water. The n-butanol extract exhibited the highest inhibitory activity against ACE, with an IC50 value of 48.82 µg/mL. The ethyl acetate extract demonstrated the highest effectiveness against α-amylase and α-glucosidase, with IC50 values of 446.15 µg/mL and 165.88 µg/mL, respectively. The n-butanol extract showed the highest DPP-IV inhibition, with an IC50 value of 139.86 µg/mL. These findings suggest that extracts from Barringtonia acutangula leaves could be beneficial in managing high blood pressure and diabetes.
