Priestia flexa—A Phosphate-Solubilizing and IAA-Producing Actinobacterium from the Rhizosphere of Oryza sativa Showing In Vitro Antagonism Against Bipolaris oryzae

To adopt eco-friendly and inexpensive alternate disease management strategies, increasing the use of actinomycetes as biofertilizers and biocontrol agents provides an alternative way to disease control. Considering the importance of using such beneficial microbes as plan growth promoters/biocontrol agents, the present study was undertaken with the following objectives giving special emphasis on the search for potential actinobacteria from the soil of rice fields having in vitro plant growth promoting (PGP) traits, potentiality as biocontrol agents. Soil samples were collected from different fields of Oryza sativa of the District Uttar Dinajpur. One potent actinobacterium, RAC-05, obtained from the rhizophere soil of Oryza sativa was found to be a phosphate solubilizer and IAA producer. RAC-05 showed phosphate-solubilizing activity by producing a clear zone on the Pikovskaya’s (PKV) medium and was positive in IAA production and catalase test. The scanning electron microscopic image of the actinobacterial isolate was taken. RAC-05 showed resistance to antibiotic chloramphenicol (30 μg) and sensitivity to ampicillin (10 μg). Actinobacterium was identified morphologically and by molecular identification as Priestia flexa (OP541862) through the submission of 16S renal fragment sequence to NCBI GenBank. Leaf pathogen-ORY-LD-01 isolate was collected from diseased leaf samples showing the symptoms of brown spot of Oryza sativa from the location of Dudhanda, West Bengal.18SrDNA fragment sequence of strain ORYLD-01 was deposited on GenBank with accession number OP541600. The fungus was identified as Bipolaris oryzae. P. flexa showed in vitro antagonism against B. oryzae. P. flexa also checked the growth of the tested pathogen significantly. The tested fungus was inhibited to some degree, and the percentage inhibition was 80.95% by P. flexa.
