Commercialized ADCs: A Market Snapshot for Exemplification

ADCs enable targeted delivery of cytotoxic payloads to cancer cells while minimizing systemic side effects. This review covers the various components of ADCs—the monoclonal antibody, linker, and cytotoxic drug payload—and the role each plays in the mechanism of action. It summarizes the 7 ADCs currently approved by FDA for cancer treatment, including their applications, pharmacokinetics, clinical efficacy and safety based on trial data. With more than 60 ADCs in clinical development, ongoing research continues to explore new disease indications, combination therapies, next-generation modifications to overcome limitations of early generation ADCs. Overall, ADCs represent an innovative therapeutic approach, using engineered antibodies to precisely target tumors with potent chemotherapies and expand the arsenal of oncology treatments.
