Novel Methods for Exosome Isolation Based on Precipitation and Chromatography

Following explosive growth in mass spectrometry and associated technology for comprehensive proteome characterization, the post-genome era presented new opportunities to discover predictive or diagnostic molecular disease biomarkersBiomarkers. To simplify sample collection, enhance early-stage detection of low abundance proteins, extracellular vesicles (EVs) present the opportune “liquid biopsy”, whose protein cargo reflects the molecular dynamics inherent to the source “diseased” tissue. Despite availability of multiple standardized protocols for EV isolation, new strategies continue to evolve to permit rapid and robust collection of EVs at high recovery and purity. This chapter focuses on approaches to isolate exosomes, based on precipitationPrecipitation or chromatographic separation, offering the benefits of being easily implemented with minimal specialized equipment while also lending scalability and high throughput. Several biologically derived or inorganic materials-based approaches have been exploited to probe selective exosome binding and capture, enabling their isolate from contaminant proteins present in biological fluids. These techniques provide promising opportunities for proteomicProteomics examination of highly enriched samples and address the needs of improved detection technologies for disease biomarkerBiomarkers discovery.
