Acokanthera schimperi is a plant known for its medicinal properties, and its leaves hold potential as a source of bioactive compounds with significant health benefits. The objective of this study was to develop reliable extraction techniques for the effective extraction of the active components from A. schimperi plant leaves. The powder from these plant leaves was extracted by maceration, Soxhlet, and sonication extraction methods, and solvents like ethanol, methanol, and distilled water were used in succession. The antioxidant activity of crude extracts was determined using a 1-diphenyl-2-picrylhydrazyl (DPPH) assay. It was observed that the sonication extraction method using methanol as the solvent resulted in the highest extract yield of 37.01 ± 0.15%, followed by Soxhlet extraction using an ethanol solvent and a yield of 29.55 ± 0.18%. The percentage radical scavenging activity (RSA) determined by the half-maximum inhibitory concentration (IC50) value was 131.08 ± 0.09 μg/ml. The data obtained in this experimental study suggest that the methanolic leaf extract of A. schimperi plant leaves possesses significant secondary metabolites that may have antioxidant properties. Consequently, these plant leaves offer a practical supply of organic antioxidants for use in pharmacological and industrial applications.
